It really depends on the friend and the girlfriend/cheater. I was in a situation where this girl I dated cheated on me (though she'll never admit to it). Friends tried to tell me about it, but I didn't believe them because I knew that the friend(s) hated the girl I was with. Thus their credibility made them liars in my eye.

This is why I would really prefer the cheater to tell me. It takes away the middle man/woman and absolves a friend from getting involved and making matters worse. However if the cheater doesn't confess, I'd want a good friend to call her out. Not the sort of thing where the friend drags you aside and says...dude, she's cheating on you. No, I'd want the friend to be smart. The friend should get as much information as possible. Then wait until the right moment when all three people (or more, depending) are together, and BAM! Expose her in front of everyone. Though that could have some seriously bad repercussions.
